The New York Jets and the New York Knicks were set to meet in a playoff game on Sunday, but the Giants beat the Seahawks.
In the first quarter, the Giants led 24-13, but Seattle’s Marshawn Lynch caught a pass and ran for a touchdown.
The score remained tied until the fourth quarter, when Manning had a big game on the ground for the first time since Week 2.
Manning was named the NFL’s Defensive Player of the Week, after posting a league-leading six sacks.
He became just the second player in league history to record five sacks in a game, joining Hall of Famers Deion Sanders and Jim Brown.
On Sunday, Manning went 6-for-7 for a career-high 153 yards and two touchdowns, as the Giants won the game for the third time in four days.
It was the first win for the Jets since they beat the Buffalo Bills 29-27 on Nov. 15.
Seahawks QB Russell Wilson and Broncos QB Trevor Siemian combined for just 2 for 14 passing.
“We just got to keep moving forward and keep believing in ourselves and we’ll be fine,” Manning said after the game.
“We just have to continue to push through it and continue to have faith in ourselves.”
Siemian also had two touchdown passes and threw for two more.
New York had to settle for a 17-16 win over the New Orleans Saints on Sunday.
After a late fourth-quarter touchdown, Manning had two sacks and a fumble recovery.
Peyton Manning has won 11 consecutive games in the Super Bowl for the New England Patriots.
